Wechselspiel von integrinvermittelter Adhäsion und MMP14-katalysierter Proteolyse in matrixdurchbrechenden Invadopodien von Tumorzellen

    Die molekulare Interaktion von b1 Integrinen und MMP14 wird in Invadopodien von Melanomzellen auf proteinchemischer und zellulärer Ebene aufgeklärt. Um die funktionelle Interaktion zu untersuchen, wird die integrinabhängige Lokalisierung und Aktivität von MMP14 in Invadopodien analysiert. Die Inhibition der Melanominvasion durch die individuelle Hemmung von MMP14 wird mit dem neuen Ansatz verglichen, Invadopodien durch bifunktionelle Integrin- und MMP14-gerichtete (BIMT) Moleküle auf zellulärer Ebene zu hemmen. Translational werden die BIMTs auf ihr Potenzial geprüft, Melanominvasion im Hühnchen-CAM-Test und im Mauslungentest zu inhibieren.
