While an infection with seasonal Influenza A virus (IAV) is accompanied by mild symptoms, and a balanced antiviral response, an infection with highly pathogenic Influenza of avian origin (HPAIV) can lead to a systemic infection and death caused by an imbalanced inflammation. However, the exact reasons behind the excessive host inflammatory response are still poorly understood.  Presumably Protein Kinase R (PKR) acts as a RIG-I independent viral RNA sensor in HPAIV, leading to the activation of a down streaming cascade involving p38 and MSK1 resulting in the phosphorylation of TRIM28. This then causes an increased production of proinflammatory cytokines. Since PKR acts as a central hub for detecting cellular stress and displays a lot of up- and downstream interaction partners it is critical to specify these interactions during infection to better understand PKRs role in excessive host inflammatory response. Therefore, the interactome of PKR will be investigated by using proximity labelling approach with the TurboID biotin ligase and Mass spectrometry over the course of IAV and HPAIV infections in lung epithelia cell lines.

Cell culture, infection studies with IAV, HPAIV and SARS-CoV-2 (BSL-2, BSL-3), Western blot, quantitative RT-PCR, Plaque assay, cloning, inhibitor treatment, mass spectrometry, proteomics
