Mechanismen und Konsequenzen von Immunzellwechselwirkungen mit Endothelzellen und Perizyten an der Blut-Hirn-Schranke

    Unser Projekt befasst sich mit den Interaktionen zwischen Immunzellen und zellulären Bestandteilen der Blut-Hirn-Schranke (BHS). Wir untersuchen Interaktionen zwischen humanen Immunzellen mit autologen Hirnendothelzellen generiert aus iPS-Zellen von Patienten mit verschiedenen neuroinflammatorischen Erkrankungen. Zusätzlich haben wir ein humanisiertes Mausmodell etabliert, um die Mechanismen der ZNS Infiltration und Läsionsentstehung nach Transfer von humanen Immunzellen zu untersuchen. Im zweiten Teil untersuchen wir die Rolle von Perizyten für die Integrität der BHS, die Entstehung chronischer Entzündung und Demyelinisierung in verschiedenen Mausmodellen.
